United boss Ole Gunnar Solskjaer was hoping for a home fixture after watching his in-form team brush aside Chelsea at Stamford Bridge.
But he has been forced to settle with a tricky clash against Nuno Espirito Santo's talented team at the Molineux, while City will travel to the Liberty stadium in south Wales.
The quarter-final matches will be played from March 15-18 with a trip to Wembley the reward for teams who progress into the last four.
United, 12 time winners of the competition, are gunning for their first FA Cup since 2016.
City will fancy their chances of progressing against the Championship outfit with Guardiola's side still in the hunt for the quadruple.
Watford are yet to win a major piece of silverware in their history having finished runners-up in the FA Cup in 1984.
Their opponents Palace have also finished runners-up on two occasions including in 2016 when they lost out to Manchester United.
Millwall famously reached the final as a Championship side in 2004 while Brighton also finished runners-up in 1983.
