Gareth Southgate put the Three Lions squad through their paces, with a win enough to seal qualification, with Alli particularly enjoying himself.
The Tottenham star, 21, was seen jokingly kneeing Jake Livermore in the ribs.
Former club team-mates Daniel Sturridge and Raheem Sterling were also spotted sharing a joke during Wednesday's session.
Alli's Spurs team-mate Harry Kane meanwhile - who has been named England captain for the game - looked typically focused as he looks to continue his fine form from recent weeks.
After a dry August, Kane scored 13 times in all competitions in September, including a perfect hat-trick away to APOEL in the Champions League.
The England star also scored twice in games against Huddersfield, West Ham, Borussia Dortmund, Everton and Malta - in the World Cup qualifier on September 1.
Southgate's Three Lions go into the game five points clear of Slovakia in the top spot of Group F - with six wins from their eight games.
However, the Slovakians sit just ONE point ahead of both Slovenia and Scotland - with all three clubs going into their penultimate group fixture with everything to play for.
England on the other hand have a much simpler task, knowing they need just one point to all-but qualify.

A win in either of their two matches would seal the deal.
On the home front, Scotland go up firstly against Slovakia - in their biggest game of the group stage so far, given the situation - before travelling away to Slovenia in their final game.
Scotland take on Slovakia on Thursday at 7.45pm at Hampden Park.
