Less than 24 hours after crashing out of the doubles along with fellow Brit Katie Swan, Boulter's singles campaign was over.
She made it hard work for herself - falling 3-0 behind in the first set before fighting back, eventually going down 6-3 in 37 minutes.
Battling Boulter then carved out an opening as she attempted to break the no18 in the second game of the second set.
The 21-year-old took it to advantage and was on the edge of breaking Osaka.
But the world no18 held on to draw level and went on to claim the second set 6-5, to win the game and progress to the third round.
Boulter's Wimbledon is still a success, though, as after the tournament she will be confirmed as the new British no.2 - leapfrogging Heather Watson.
