The Spider-Man and X-Men creator, 95, was taken in suffering shortness of breath and an irregular heartbeat, TMZ report.
Stan is a pioneer in the comic industry, having worked as a writer, editor, film exec and publisher.
He was formerly editor-in-chief of Marvel Comics and has made a cameo in every single Marvel film to date.
He co-created legendary characters such as Spider-Man, the Hulk, Doctor Strange, the Fantastic Four, Daredevil, Black Panther and the X-Men.
Lee was taken to Cedars-Sinai Medical Centre in LA on Wednesday night.
A spokesperson for Stan said: "He is doing well and feeling good, he is staying there for a few days for some check ups as a safety precaution."
He cancelled two major convention appearances last year through illness, but never revealed the specifics.
Last month, we revealed how Stan has been accused of sexually assaulting nurses that come to his Hollywood home to care for him.
His legal team categorically denied the allegations.
Stan Lee's lawyer Tom Lallas told Sun Online the allegations against the Marvel creator were "false and despicable".
He said: "Mr Lee categorically denies these false and despicable allegations and he fully intends to fight to protect his stellar good name and impeccable character.
